TY - JOUR AU - U-Kong, Warut AU - Chundet, Ruttaporn AU - Chandet, Nopakarn PY - 2017 TI - การศึกษาการส่งถ่ายยีน gus เข้าสู่ปทุมมากระถางด้วยเชื้อ Agrobacterium tumefaciens JF - Naresuan University Journal: Science and Technology (NUJST); Vol 25 No 4 (2017): October - December 2017 KW - potted curcuma, gus gene, retarded shoots, Agrobacterium tumefaciens N2 -      งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาการส่งถ่ายยีน gus เข้าสู่ปทุมมากระถางโดยใช้แบคทีเรีย Agrobacterium tumefaciens ในการส่งถ่ายยีน เมื่อนำยอดปทุมมากระถางมาเลี้ยงบนอาหารแข็ง MS ที่เติม IAA ความเข้มข้น 0.1 มิลลิกรัมต่อลิตร TDZ ความเข้มข้น 0.5 มิลลิกรัมต่อลิตร และ IMA ความเข้มข้น 4.0 มิลลิกรัมต่อลิตร พบว่าสามารถชักนำให้เกิด retarded shoots ได้ 100 เปอร์เซ็นต์ จากการศึกษาผลของสารปฏิชีวนะต่อการเจริญเติบโตของ retarded shoots โดยวาง retarded shoots บนอาหารที่เติมสารปฏิชีวนะที่มีความเข้มข้นต่างๆ กัน เป็นเวลา 6 สัปดาห์ พบว่าความเข้มข้นที่เหมาะสมของสารปฏิชีวนะซีโพแทกซีมและกานามัยซินที่สามารถยับยั้งการเจริญเติบโตของเชื้ออะโกรแบคทีเรียและยับยั้งการเจริญเติบโตของ retarded shoots ที่ไม่ได้รับการส่งถ่ายยีนคือ 250 และ 100 มิลลิกรัมต่อลิตร ตามลำดับ เมื่อทำการส่งถ่ายยีนโดยนำ retarded shoots ประทุมมากระถางมาเขย่าร่วมกับแบคทีเรีย A. tumefaciens สายพันธุ์ LBA4404 ซึ่งมีพาหะลูกผสม pStart ที่มียีน gus แล้วนำมาวิเคราะห์การแสดงออกของยีนด้วยวิธี GUS histochemical assay ผลการทดลองพบว่าเนื้อเยื่อที่ได้รับการส่งถ่ายยีนโดยการเขย่าร่วมกับแบคทีเรียเป็นเวลา 30 นาที จะมีการแสดงออกของยีนสูงสุดถึง 51.61% คำสำคัญ: ปทุมมากระถาง ยีน gus retarded shoots Agrobacterium tumefaciens      The objective of this study was to transfer gus gene into potted curcuma via Agrobacterium tumefaciens transformation. The shoots of potted curcuma were cultured on MS solid medium supplemented with IAA (0.1 mg/l), TDZ (0.5 mg/l) and IMA (4.0 mg/l). A hundred percent of retarded shoots induction was observed. The retarded shoots were then cultured on the media supplemented with different concentration of antibiotics for 6 weeks. The appropriate concentration of cefotaxime and kanamycin that could inhibit the growth of Agrobacterium and the induction of non-transgenic shoots were 250 mg/l and 100 mg/l, respectively. The transformation was subsequently performed by shaking incubation of potted curcuma retarded shoots with A. tumefaciens LBA4404 carrying gus recombinant vector pStart. The expression of gus gene in transgenic shoots was investigated by GUS histochemical assay. Results revealed that the expression of gus gene was highest (51.61%) in transgenic shoots incubated with Agrobacterium for 30 min.
